Historically, virginity, at least in a woman, was sought after when a couple were due to marry and probably start a family because it would mean the woman would not already be pregnant with another man's child. That attitude would have emerged at a time before modern contraception and when it was usual for the man to work to support the woman who then mostly raised the children.
Given the link with pregnancy in the above, the only thing that should count as losing one's virginity is penis in vagina sex. Of course, lesbians may then say that isn't fair and that dildo in vagina should count and gay guys could complain and say that penis in anus should count. Really, in a same sex situation, or with effective contraception, it is an outdated concept anyway. -

Assuming by "checking it" you mean checking that the hymen is intact. That is not a reliable indicator, though. The hymen can be broken by things other than sex. That also gives rise to girls seeking hymen repair. The whole things seems like a silly obsession and rather pointless. -
